Role of Type I IFNs in the in Vitro Attenuation of Live, Temperature-Sensitive Vaccine Strains of Human Respiratory Syncytial Virus

AbstractThe contributions of type I interferons (IFNs) to the in vitro attenuation of three temperature-sensitive (Ts) subgroup A and one subgroup B deletion mutant RSV strains were evaluated. The ability of these vaccine viruses to induce IFNs at their permissive and restrictive temperatures and their sensitivity to the antiviral effects of exogenous I IFNs were tested in human lung epithelial A549 cells. Our results show that the highly attenuated and immunogenic subgroup A vaccine strain Ts1C produced higher levels of IFN-β than its parent RSS-2 or two related strains, Ts1A and Ts1B, at their permissive temperature. Growth of RSV-infected A549 cultures at restrictive temperatures or prior UV inactivation of the virus abolished the observed induction of IFN-β, suggesting a strict requirement of viral replication for cellular IFN induction. The enhanced induction of IFN-β by the highly immunogenic Ts1C at permissive temperature may be an advantageous characteristic of a live intranasal vaccine candidate. The subgroup B strain RSV B1 and its mutant cp-52 (with SH and G gene deletions) both induced similar but low levels of IFN-β. Hence the observed overattenuation of cp-52 in human infants is probably not due to enhanced IFN induction during its replication in the host. The ability of cp-52, which does not express the SH and G proteins, to induce IFN-β levels similar to those of its parent strain suggests that these viral proteins may not have a role in the induction of IFN-β in the host. In addition, both subgroup A and B mutants and their respective parent strains were similarly resistant to the antiviral effects of exogenous IFN-α or -β. Therefore, increased sensitivity of the mutants to IFNs does not seem to contribute to their attenuation

A method was developed through leaf as explant of Dysophylla myosuroides (Roth.) Benth. for multiple shoot regeneration. The effect of Auxins (IAA, NAA, IBA) and Cytokinins (BA) were studied on shoot regeneration in clulture. On MS half strength medium light green compact calli were formed with 0.1 mg l-1 NAA. The cultures produced 62.57 + 0.04 shoots on half strength medium with 0.5 mg l-1 BA and 0.01 mg l-1 NAA and also maximum mean length (0.52 + 0.02) of shoots were achieved. In vitro produced shoots rooted on half strength MS medium with 1 mg l-1 IBA. The in vitro regenerated plantlets were successfully acclimatized in paper cups containing vermiculate, then transferred to green house. Hardened plants were transplanted in to sand and soil (1 : 1)

A transform of complementary aspects with applications to entropic uncertainty relations

Even though mutually unbiased bases and entropic uncertainty relations play an important role in quantum cryptographic protocols they remain ill understood. Here, we construct special sets of up to 2n+1 mutually unbiased bases (MUBs) in dimension d=2^n which have particularly beautiful symmetry properties derived from the Clifford algebra. More precisely, we show that there exists a unitary transformation that cyclically permutes such bases. This unitary can be understood as a generalization of the Fourier transform, which exchanges two MUBs, to multiple complementary aspects. We proceed to prove a lower bound for min-entropic entropic uncertainty relations for any set of MUBs, and show that symmetry plays a central role in obtaining tight bounds. For example, we obtain for the first time a tight bound for four MUBs in dimension d=4, which is attained by an eigenstate of our complementarity transform. Finally, we discuss the relation to other symmetries obtained by transformations in discrete phase space, and note that the extrema of discrete Wigner functions are directly related to min-entropic uncertainty relations for MUBs.Comment: 16 pages, 2 figures, v2: published version, clarified ref [30

Introduction. Photovoltaic (PV) systems gained immense attraction in the recent years since it produces electricity without causing environmental pollution through direct conversion of solar irradiance into electricity. Solar PV panels produce DC power. The magnitude of this DC power varies with temperature and irradiance of the sun rays. The DC supply from solar panels can be regulated using DC-DC converter and then can further be converted into the desired AC voltage by means of a voltage source inverter before being fed to an induction motor (IM). The speed and torque of an IM, fed from PV arrays, can vary due to the variation in the output power of the panels. Goal of this work is to improve the dynamic performance and reduce the torque ripple of Cuk converter-inverter fed IM drive system. The novelty of the current work proposes interleaved Cuk converter between solar PV DC source and the inverter. Purpose. To provide continuous current using an interleaved Cuk converter to the IM drive and in turn to reduce the torque ripple in IM. Methodology. Introduced an interleaved Cuk converter which is a blend of Cuk converters connected in parallel with each other between solar PV arrays and IM drive system. Originality. Simulation results are obtained for Cuk converter and interleaved Cuk converter fed IM drive by means of MATLAB. The hardware setup for the same IM systems is developed. Practical value. Simulation and hardware results are coincided with each other and it is subject from the simulation and hardware results that the interleaved Cuk converter-inverter fed IM system produced results superior than the Cuk converter inverter fed IM drive system.
